Kwabena Kesse built a football team in Kumasi and named it after himself, Kessben F.C. It remained a Kumasi side for eight years, until the businessman Moses Armah Parker bought the club in 2010 for $600,000 and moved it south-west to Tarkwa, a mining town in Ghana's Western Region. The following January it was renamed Medeama Sporting Club, and the mauve-and-yellow kit that gave the side its nickname, the Mauve and Yellows, became a fixture at T&A Stadium, a 10,400-seat ground shared with the neighbouring town of Abosso.

The first trophy arrived in 2013, when Philip Anoba's goal beat Asante Kotoko in the FA Cup final; two years later Medeama beat the same opponent again, 2-1. The league title took longer to land — Medeama won the Ghana Premier League in the 2022–23 season and repeated it in 2025–26. Joseph Tetteh Zutah has worn the captain's armband since 2017, through both championships.

Two fixtures outweigh the rest of the calendar: the meetings with Karela United are known as the Nzema Derby, and those with FC Samartex 1996 as the Western Derby — the two rivalries closest to home since Medeama left Kumasi for Tarkwa.
