Ayo Oke made her senior debut for the United States on 27 January 2026 against Chile, starting the match and setting up Croix Bethune's opening goal in a 5-0 win, replacing Avery Patterson.

Her path to that debut ran through the American youth ranks, where she progressed through the U-15 to U-20 levels and was part of the squad that won the 2022 CONCACAF Women's U-20 Championship before playing at the FIFA U-20 Women's World Cup that year. She later added three caps for the U-23 side.

At California, she was named to the Pac-12 All-Freshman Team in 2021 and earned Second-team All-Pac-12 honours in 2022. She then transferred to UCLA, where she helped the Bruins win the Pac-12 Conference title in 2023 and the Big Ten Tournament in 2024, adding a second All-Pac-12 selection along the way.

She turned professional with Pachuca in Liga MX Femenil in January 2025, a move that brought her the Clausura 2025 title and the Campeón de Campeonas trophy after 31 league appearances and a playoff goal. A year later, Denver Summit FC signed her from Pachuca on a three-year contract, in a transfer reported at around $450,000 — described at the time as the highest fee paid for an American player entering the NWSL. She made her league debut for Denver on 14 March 2026 against Bay FC.
