Uni Souza lost the 2024 final of the Campeonato Carioca Série C to Niteroiense on penalties. The club had played professionally for three seasons by then.

Its beginnings were far more casual. Brothers Cosmo and João Souza started it in Penha, in the north of Rio de Janeiro, and called it Dá-lhe Souza Futebol Clube. That name lasted until 1990, when the club affiliated with the Rio de Janeiro state federation (FERJ) and was made to change it. It became Uni Souza. The black and white kit and the nickname Alvinegro da Penha stayed, and the mascot is an eagle.

For most of its life the club stayed in amateur football, in the Campeonato da Beira da Linha, a competition run around the Penha Circular area. The Torneio Início of that league, the opening tournament of the season, was its first trophy, in 1992. Falcão and União Central are the sides it met in finals in those years. In 2012 the under-15s won the Copa Rio Amador da Capital.

Professionalisation came in 2021 under president Gilberto Nascimento, with entry into the Série C. The club's home is the Estádio Moça Bonita, officially the Estádio Proletário Guilherme da Silveira Filho. It stands in Bangu, and holds 9,024.
