The club itself came together on 3 January 1915, a merger of Lanús United and Club Social El Progreso, named for Anacarsis Lanús, the Frenchman who founded the town in 1854. What followed was not a straight climb. Promotion to the Primera came in 1919, then in 1934 the Argentine federation forced a temporary fusion with neighbouring Talleres de Remedios de Escalada, and in 1949 a relegation still remembered as scandalous. The 1950s brought the closest thing to a golden age before the modern trophy count started: a team nicknamed the Globetrotters finished runner-up in the 1956 championship.

The 2007 Torneo Apertura was the club's first title of the professional era; 2016 brought the league championship along with the Copa Bicentenario, won against Racing, and a 3-0 rout of River Plate for the Supercopa Argentina. Further afield, Lanús beat Atlético Mineiro on penalties to lift the 2025 Copa Sudamericana, then went to the Maracanã in 2026 and won there too, overturning Flamengo across two legs for the Recopa Sudamericana.

Diego Milito wore the shirt between 2005 and 2008 before winning the Champions League with Internazionale; Lionel Scaloni, now Argentina's head coach, played there from 1999 to 2001. The fiercest fixture remains the Clásico del Sur against Banfield.
