Villa Acassuso had no club of its own until a group of amateur footballers formed one on 7 September 1922, calling it Villa Acassuso Football Club. A donation from Ernesto de las Carreras paid for its first ground, and when Argentine football split into rival associations in the 1930s, Acassuso found itself, briefly and unhappily, in the country's top flight — a spell that ended in relegation back to the lower divisions.

The club has spent most of its history there. It won Primera C twice, in 1937 and again in 2006–07, and Primera D twice, in 1971 and 2000–01. The 1946 expropriation of its ground by the municipality left it without a home until San Isidro handed over land on Calle Santa Rita in 1977; Estadio La Quema opened there in 1983 and still holds only 1,500 people. The nickname Los Quemeros comes straight from the ground's name.

Rodrigo Alonso holds the appearance record, 233 games and 21 goals between 2006 and 2014. Brian Romero came through the same ranks from 2012, moved on to River Plate and the national team, and later returned to public attention after being diagnosed with rheumatoid arthritis.
