Mirassol Esporte Clube took shape in the interior of São Paulo state on 9 November 1925. A 1964 merger with Grêmio Recreação, Esporte e Cultura briefly reshaped the club into Mirassol Atlético Clube, with white and blue replacing the original colours; the arrangement lasted until 1981, when the merger was undone and the club returned to its old name and its yellow and green.

Matches are played at the Estádio Municipal José Maria de Campos Maia, known locally as Maião, which opened on 3 March 1983 with a 2-1 win over Jalesense. A safety report filed by the São Paulo state football federation in November 2022 put its capacity at 15,023.

For most of its history Mirassol was a regional name, its clearest early mark a Campeonato Paulista Série A3 title in 1997. What changed came later and quickly: promotion through Série D in 2020, the Série C title in 2022 — the year that added a green star to the club crest — and a runners-up finish in Série B in 2024. Negueba was the attacking presence through that climb from the third tier to the first. Camilo, who scored 28 goals for the club and was part of the 2022 Série C-winning side.

The academy has done its own work in parallel: Luiz Araújo moved on to Flamengo and later to clubs in Europe, and in 2026 two more products of the youth system, the defender Ramirês and the midfielder Cristian Araújo, followed the same path to Flamengo. Locally the rivalry that matters most is with Linense, played as the Clássico Caipira; the meeting with Novorizontino carries its own name, the Clássico Tião.
